The t(15;17)(q22;q21) translocation is commonly associated with Acute Promyelocytic Leukemia (APL), a subtype of Acute Myeloid Leukemia (AML) classified as M3 morphology. APL accounts for approximately 10-15% of AML cases. This translocation results from the fusion of the PML gene on chromosome 15 and the RARa (Retinoic Acid Receptor alpha) gene on chromosome 17. Breakpoints in these genes occur in different regions, producing specific PML-RARa isoforms named bcr1, bcr2, and bcr3. The fusion protein blocks differentiation of hematopoietic stem cells, arresting them at the promyelocyte stage. Treatment with ATRA (All-Trans Retinoic Acid) is highly effective in APL by promoting differentiation.
