Clinical Significance
The p63 protein helps distinguish prostatic adenocarcinoma from benign prostatic hyperplasia (BPH).
For example, malignant glands lack p63 expression, while basal cells in BPH show strong positivity.
Moreover, when combined with p40 and CK5/6, p63 supports the diagnosis of squamous cell carcinoma in the head, neck, lung, and breast.
Assay Principle
During the IHC procedure, the primary p63 antibody binds directly to its nuclear antigen.
Next, the secondary HRP-conjugated antibody attaches to the primary antibody and strengthens the signal.
Finally, the DAB chromogen reacts with this complex and produces a clear brown nuclear stain.
Therefore, the staining confirms the presence of the p63 antigen.
