🧪 MDM2 Antibody (Immunohistochemistry)
The MDM2 Antibody is a key immunohistochemical marker used for identifying well-differentiated and dedifferentiated liposarcomas as well as other tumors with MDM2 gene amplification.
MDM2 is a negative regulator of the tumor suppressor p53, involved in controlling the cell cycle and inhibiting apoptosis, thereby contributing to tumor progression and survival.
Overexpression or gene amplification of MDM2 is observed in a variety of neoplasms, including soft tissue sarcomas, osteosarcomas, and certain epithelial tumors.
🔬 Features and Benefits
-
Specific marker for liposarcoma: Differentiates liposarcoma from benign lipoma
-
Distinct nuclear staining: Brown nuclear coloration in positive tumor cells
-
Broad diagnostic utility: Effective for soft tissue and bone tumor evaluation
-
Compatible with standard IHC protocols
-
Stable performance: Shelf life of 18 months at 2–8 °C
⚙️ Principle of the Assay
During the IHC process, MDM2 antibody binds to the nuclear antigen within tumor cells. The HRP-conjugated secondary antibody binds to the primary antibody to form the antigen–antibody–HRP complex. Upon addition of DAB chromogen, a brown insoluble stain appears in the nuclei of positive cells, confirming MDM2 expression.
💡 Diagnostic Applications
-
Detection of Well-differentiated and Dedifferentiated Liposarcoma
-
Differentiation between liposarcoma and benign lipoma
-
Assessment of MDM2 gene amplification in bone and mesenchymal tumors
-
Research related to p53 signaling and apoptosis regulation
