STAT6 is a 94 kDa nuclear transcription factor activated through IL-4 and IL-13 signaling.
As a result, abnormal nuclear expression of STAT6—caused by the NAB2–STAT6 fusion gene—acts as a distinctive diagnostic feature.
In addition, strong nuclear STAT6 staining is considered a sensitive and specific indicator of SFT.
Therefore, it helps pathologists differentiate SFT from other fibroblastic and mesenchymal tumors with greater accuracy.

⚙️ Principle of the Assay
During the IHC procedure, the primary STAT6 antibody binds directly to its nuclear antigen.
Next, the HRP-conjugated secondary antibody attaches to the primary antibody.
Finally, the DAB chromogen reacts with this complex and produces a brown nuclear stain.
Consequently, this staining confirms STAT6 expression in tumor cells.
