Calretinin Antibody – Product of Talent Biomedical
Pathologists use the Calretinin Antibody in Immunohistochemistry (IHC) to detect Calretinin,
a 29 kDa calcium-binding protein. Calretinin supports calcium signaling and contributes to cellular
differentiation. As a result, it appears in normal and neoplastic mesothelial cells and
also in Leydig cells.
In addition, this antibody helps differentiate malignant mesothelioma from adenocarcinoma,
especially in lung and pleural biopsies. Therefore, Calretinin is considered one of the most
sensitive and specific markers for mesothelioma.

🔬 Clinical Significance of Calretinin
Calretinin serves as a key diagnostic marker for mesothelial differentiation.
Moreover, its combined nuclear and cytoplasmic staining strongly supports a diagnosis of
mesothelioma. In contrast, adenocarcinomas usually remain negative for this marker.
Therefore, including Calretinin in an IHC diagnostic panel significantly improves the
accuracy of distinguishing mesothelioma from adenocarcinoma.
🧫 Principle of the Assay
During IHC staining, the primary Calretinin antibody binds directly to its cytoplasmic and nuclear antigens.
Next, the HRP-conjugated secondary antibody attaches to the primary antibody.
Finally, after adding the DAB chromogen, a brown insoluble stain appears.
As a result, the staining confirms the presence of the Calretinin antigen.
