Clinical Significance
First, CEA acts as an oncofetal glycoprotein of about 200 kDa.
You normally find it on the surface of gastrointestinal epithelial cells.
However, malignant tumors show a much higher level of CEA expression.
In addition, the staining pattern often appears as a linear signal along the apical membrane of glandular cells.
Therefore, CEA serves as a useful marker for diagnosis, tumor staging, and treatment planning in cancer patients.
Assay Principle
During the IHC procedure, the primary CEA antibody binds directly to the CEA antigen in the tissue section.
Next, a secondary HRP-conjugated antibody attaches to the primary antibody and forms a stable complex.
Finally, the DAB chromogen reacts with this complex and produces a brown, insoluble signal that marks CEA-positive areas.
