β-Catenin is a 92 kDa cytoplasmic and nuclear glycoprotein that plays dual roles in cell adhesion (via E-Cadherin) and transcriptional regulation.

In normal cells, β-Catenin is localized to the cell membrane, but abnormal activation or mutation of the Wnt pathway results in nuclear translocation, which activates genes related to tumor proliferation.
Thus, β-Catenin IHC staining is crucial for diagnosing Desmoid Tumor, Hepatocellular Carcinoma, Colorectal Carcinoma, and Medulloblastoma.
