Pathologists use the CA 125 Antibody to detect Cancer Antigen 125 (CA 125) in tissue samples.
CA 125 belongs to the MUC16 mucin family and appears as a membrane-associated glycoprotein.
As a result, this marker plays a key role in identifying ovarian serous carcinomas and epithelioid mesotheliomas.

In addition, CA 125 normally appears in endometrial, fallopian tube, and cervical epithelium.
However, it does not appear in normal ovarian tissue.
Therefore, strong CA 125 staining helps distinguish epithelial ovarian carcinoma from other malignancies.

Clinical Significance

CA 125 acts as one of the most established tumor markers for ovarian carcinoma.
Moreover, its overexpression appears in epithelial ovarian carcinomas, mesotheliomas, and some endometrial tumors.
Consequently, evaluating CA 125 expression helps determine tumor origin and distinguish primary tumors from metastases.

Assay Principle

During the IHC procedure, the primary CA 125 antibody binds directly to its membrane and cytoplasmic antigen.
Next, a secondary HRP-conjugated antibody attaches to the primary antibody.
Finally, the DAB chromogen reacts with the complex and produces a brown stain at the antigen site.
This staining confirms CA 125 expression in the tissue.
