The PMS2 (Postmeiotic Segregation Increased 2) Antibody is one of the four key markers used to evaluate the Mismatch Repair (MMR) system for detecting Microsatellite Instability (MSI), particularly in colorectal and endometrial carcinomas.

PMS2 protein, along with MLH1, MSH2, and MSH6, plays an essential role in correcting DNA replication errors.
Loss of PMS2 nuclear staining by IHC is a major indicator of defective MMR genes, often associated with Lynch Syndrome (Hereditary Nonpolyposis Colorectal Cancer).
